Outline of Annual Research Achievements

1.ヒストン蛋白質修飾の検討
若年群と加齢群のマウスより側頭骨を摘出し、ホモジェナイズ後、ヒストン蛋白質定量キットを用い、若年群と加齢群のヒストン蛋白質修飾の変化を検討中であるが、現時点で有意差は明らかではなかった。
パラフィン包埋した側頭骨から切片を作成し、抗ヒストン蛋白質抗体を用い、蛍光染色を行った。その結果、剥年郡ではH3k9およびH4k8のアセチル化およびH3k27のメチル化、加齢群ではH3k9のメチル化を認めた。これらの結果を踏まえ、ヒストン蛋白質修飾定量法を検討中である。
2.DNAメチル化の検討
側頭骨よりDNAを抽出した。DNA量は加齢群で若年群に比べ、約50％に減少した。その後、バイサルファイト処理を行った。バイサルファイト未処理なDNAと処理済みのDNAに対応したプライマーを作成した。qPCRを行い、検討中である。

マウス側頭骨より採取できるDNA量は非常に少なく、5匹、10耳より採取したDNAを用い、検討中である。
