坐骨神経疼痛及びその回復過程におけるGABAの機能

Outline of Annual Research Achievements

平成27年度に続き、野生型を用いて慢性疼痛動物モデルを作製している。坐骨神経圧迫モデルでは、手術3日目に下肢運動機能が有意に低下し、1ヵ月後運動機能が回復することを確認した。損傷後の脊髄では、前角と後角ともにK-Cl-Cotransporter2(KCC2)低下した後、徐々に回復した。この結果から、坐骨神経損傷後脊髄においてGABAは興奮性に作用し、神経の再生特に、軸策の再生に関与している可能性が示唆された。共に、頚骨神経切断モデルを作製し、運動機能および感覚機能を解析した。切断後14日目まで下肢運動機能は著しく低下し、3ヶ月に掛けて一部機能回復を示した。Von Frey testでは損傷側で感覚異常が生じた。頚骨神経が支配している脊髄後角内側でのKCC2低下および回復が確認できた。この結果から、神経損傷を直接受けている脊髄のGABA関連物質の解析が可能になった。続いて、GABA関連分子欠損マウスを用いて頚骨神経切断モデルを作製し、行動評価をしている。
