Outline of Final Research Achievements |
By serial passaging of PB2 segment deficient virus (ΔPB2 virus) in PB2 stably expressing MDCK cells, we obtained 3 strains ofΔPB2 virus with high growth property. All the 3 strains possessed mutations in their NP proteins. The NP mutations caused sifting the charge to basic side. Although ΔHA virus lacked its HA segment, ΔHA virus possessed 8 genome segment in its virion. Next generation sequencing and Northern blotting analysis revealed that ΔHA virus incorporated host-derived rRNA in its virion. These findings highlight the importance of the assembly of eight RNPs in progeny virions for genome packaging.
