Outline of Final Research Achievements |
In this study, we applied tensile force to human PDL fibroblasts to analyze relationship between Scleraxis (Scx), a transcription factor for tendon/ligament cell lineages, and TGF-β/Smad signaling.Scx mRNA expression was significantly up-regulated at 3 h. We also detected activation of p-Smad3. Pretreatment with SB-431542 or SIS3 showed not only inhibition of phosphorylation of Smad3 in a dose-dependent manner but also down-regulation of Scx mRNA. In conclusion, up-regulation of Scx mRNA in response to tensile force is regulated via TGF-β/Smad signaling.
