Outline of Annual Research Achievements

歯槽骨を含む顔面頭蓋の骨格の多くは膜性骨化によって形成される。申請者らは、Rhoファミリータンパク質であるCdc42が個体レベルでの骨・軟骨形成において、軟骨内骨化での正常な軟骨細胞の肥大化や軟骨基質の石灰化、肢芽形成期での軟骨細胞の凝集およびパターニングに重要な役割を果たしていることを明らかにした。一方、Cdc42が個体レベルの膜性骨化において果たしている機能は未だ不明な点が多い。
そこで、頭蓋における間葉細胞特異的にCdc42遺伝子を欠損させたコンディショナルノックアウトマウスの表現型の解析を行ったところ、野生型マウスと比較して頭蓋縫合部の骨形成が有意に減少していたものの、細胞増殖および細胞死マーカーの発現に有意差は認められなかった。Cdc42コンディショナルノックアウトマウスの頭蓋においては、血管新生の低下とそれに伴う浮腫の形成が認められ、頭蓋縫合部の石灰化を制御するIhh、Bmp2およびBmp4の発現減少が認められた。
これらの結果から、Cdc42が頭蓋における血管新生を制御し、間接的にIhhおよびBmpシグナルを制御することにより、頭蓋骨形成において重要な役割を果たしていることが示唆された。
