Outline of Annual Research Achievements

細胞運動のモデル種である細胞性粘菌Dictyostelium discoideumでは数万個の細胞が走化性誘因物質としてサイクリック ＡＭＰ(ｃＡＭＰ)を自己分泌的に放出する。細胞全体で応答と放出のリズムが揃うことで、回転しながら外側に伝播するｃＡＭＰのらせん波が自己組織的に形成され、これに向かう方向（順方向）の走化性によって細胞が集合する。本研究課題では、単一細胞レベルのシグナル応答の動作原理と自己組織化の数理に立脚し、光による分子活性制御を用いて介入することで、cAMPの回転らせん波と細胞の動きをリアルタイムに操作し、自己組織化されるパターンがとりうる多様な表現型空間を構成的に明らかにすることを目的とした。光依存的に低分子量GタンパクＲａｓやＲａｃを活性化させるオプトジェネティクスの系を細胞性粘菌で初めて実装し、細胞変形に与える影響などを中心に、その基本動作をライブセルイメージングを中心に調べた。特に、Ｒａｓ、Ｒａｃ、ＰＩ3キナーゼのベーサル活性との関係について解析を進めた。さらに、新たにわかってきた細胞遊走の細胞間接着因子依存性に着目し、これを制御することで、細胞の運動を完全に停止させる条件、またcAMPの勾配とは直交する方向への運動を誘導させることに成功し、集団運動をともなったパターン形成を２つの基本的なモードに分解して理解できることを構成的に示した。最終年度は、これまで培ったオプトジェネティクスの系と、微小流体デバイスによる操作を組み合わせることで、集合塊におけるcAMPの分布、細胞配置と回転運動の制御をこころみた。内在の駆動系による動きが支配的であり、オプトジェネティクスによる集団動態への十分な摂動は得られなかっものの、微小チャンバーによる定量的な動態解析が進み、回転運動の制御にSCARの膜局在の制御の重要性など今後の課題が明らかになった。
