Research Abstract

本研究では、モデル高等植物のシロイヌナズナを用い、シロイヌナズナDNAマイクロアレイを用いた解析により、種々のストレス、ホルモンに応答する遺伝子等を単離、同定する。同定された遺伝子に関してプロモーター解析を行う。
約7000個のシロイヌナズナ完全長cDNAクローンを含むcDNAマイクロアレイやアジレント22Kオリゴアレイを用いて、種々のストレスやホルモン処理、種々の組織、種々の変異体やトランスジェニック植物など約300種類の異なる条件下での発現プロファイル解析を行った。論文発表したものの幾つかに関しては発現データを公開した。
アジレント22Kオリゴアレイなどを用いて、低温馴化および低温脱馴化過程での遺伝子発現プロファイル解析を行った。その結果、低温馴化過程で発現誘導される遺伝子の多くは脱馴化過程で発現が減少する事、および脱馴化過程で発現誘導される遺伝子の多くは低温馴化過程では発現誘導されないことがわかった。また、ラフィノースの合成に関与するガラクチノール合成酵素遺伝子(AtGolS3)やマルトースの合成に関与するβ-アミラーゼ遺伝子(At4g17090)などが植物の耐凍性獲得に主要な役割を果たしていることが示唆された。
また、シス・トランス因子を迅速に同定していく上で、プロモーターをカタログ化する事が重要である。これまでにマイクロアレイ解析により同定したストレス誘導性遺伝子約600個に関して、プロモーター断片のサブクローニング、プロモーター+GUS(LUC)のベクターコンストラクトの作製、アグロバクテリウムへの形質転換を行った。今後、プロモーター+GUS(LUC)のトランスジェニック植物の作製、GUS(LUC)遺伝子の発現解析、シス配列の探索を行う予定である。
