Research Abstract

プロスタグランジン(PG)F_<2α>は、子宮平滑筋の収縮作用を有する生理活性物質の1つで、陣痛促進剤として広く臨床応用されている。寄生虫の増殖とPGF..の因果関係は未解明であるが、感染した家畜の血中でPGF_<2α>が大量に生合成され流産の原因となり、Nagana病と呼ばれている。本研究では、トリパノソーマ(Trypanosoma brucei、T.cruzi)、リーシュマニア(Leishmania major)などの病原性寄生原虫が持つヒトと薬剤感受性の異なるPGF合成酵素を分子生物学的に同定し、そのX線結晶解析により寄生原虫のPG生合成系を標的としたSBDDのための構造基盤を構築することを目的とした。
アフリカ睡眠病の病原虫Trypanosoma brucei由来PGF合成酵素(TbPGFS)について、cDNAのクローニング、酵素の同定および反応性についての生化学的実験を行い、酵素の大量精製、結晶化、2.1Å分解能でのX線構造解析に成功した。本酵素は、アルドケト還元酵素(AKR)のファミリーに属し、活性中心で保存されているCatalytic tetrad(Asp47,Tyr52,Lys77,His110で構成)のいずれも必須不可欠と思われたが、singleおよびdouble変異体の作製および活性測定を行った結果、Lys77とHis110のみが反応に関与する、新たなCatalytic dyadの反応機構を提唱した。続いて、リーシュマニア症の病原虫Leishmania major由来(LmPGFS)についても同様に1.9Å分解能でのX線構造解析を行った。β-バレルの上側のループ領域で大きく異なる構造を有する他は、アミノ酸配列の相同性が61%と高いのを反映して、活性部位付近の構造もほぼ同一であり、反応機構も同一と考えられた。
一方、活性中心にフラビンを有するシャーガス病の病原虫Trypanosoma cruzi由来の酵素(TzPGFS)についても分子置換法により構造解析に成功し、構造の精密化中である。
今後、TbPGFSやLmPGFSとは異なる、ラジカルの関与する反応機構も構造から議論する予定であるが、病原虫の繁殖におけるPGF_<2α>の役割を解明する目的でこれら3種の寄生虫由来の蛋白質の構造基盤をもとに阻害剤の開発を行う予定である。
