Research Abstract

アフリカツメガエル(以下、ゼノパス)およびマウスの卵細胞をモデル実験系として、受精に伴う発生プログラムの活性化に関わる分子メカニズムの解析を行い、以下の知見を得た。ゼノパスにおいては、卵細胞膜マイクロドメイン(以下、卵ラフト)の構造と機能について解析を行い、単一細胞膜貫通型蛋白質uroplakin III(以下、UPIII)が、その細胞外ドメインによって、精子との相互作用(接着あるいは融合、あるいはその両方)、そして、細胞内ドメインが受精成立の卵細胞内シグナル伝達に、それぞれ関与する可能性を示唆する結果を得た。UPIII細胞外ドメインの具体的な機能については、精子に由来するプロテアーゼ活性によって部分的に切断されること、この切断反応を阻害する薬剤が受精阻害を引き起こすこと、といった、受精成立に必要な精子プロテアーゼ活性の卵細胞膜ターゲットとしての理解が深まった。細胞内ドメインの機能については、前年度までに既に明らかにしていたチロシンキナーゼSrcによる受精依存的なリン酸化に加え、テトラスパニン型相互作用因子であるuroplakin Ib(以下、UPIb)との協調作用によるシグナル伝達への関与が示された。すなわち、UPIIIはUPIbと共に発現されることで複合体を形成することによって、卵細胞膜および卵ラフトへの局在が可能になること、そして、UPIII/UPIb複合体はチロシンキナーゼSrcの不活性化を誘導すること、等が明らかになった。これらの結果を総合すると、ゼノパス卵の受精時に精子がプロテアーゼ活性により卵ラフトのUPIII/UPIb複合体に相互作用し、UPIIIの部分的な切断が何らかのメカニズムによって細胞内のチロシンキナーゼSrcの活性化に翻訳され、チロシンリン酸化のシグナル伝達が点火され、カルシウム経路の活性化へ導かれていく、という流れが想定される。マウスにおいては、精子に特異的に発現するリン脂質分解酵素phospholipase Cζの、受精に伴うカルシウムオシレーションという現象への関与について、特異的抗体や変異蛋白質の作成により解析した。その結果、phospholipase Cζが、受精時に卵細胞内へ導入される可能性が高いことや、もともと単一ポリペプチドである本分子の部分的な切断と断片化されたフラグメントが再会合が活性調節に関わる可能性等が示された。
