Research Abstract

我々はDNaseγが筋芽細胞の分化過程に於いて誘導されるアポトーシスのDNA断片化を触媒することを見いだした。この知見は、増殖期と分化期では細胞死の機構が違うこと、さらに分化過程にある細胞ではDNaseγを中心とするアポトーシス経路が準備され、分化異常をきたした細胞を首尾よくアポトーシスへと導き得る状態にシフトしていることを示唆している。本研究は細胞分化過程に於けるアポトーシス切り替え機構を明らかとすることを目標に行われた。
神経細胞分化過程に於いて、細胞周期を進行する未分化細胞ではCAD発現が認められるが、静止期にある分化細胞ではCADは消失しDNaseγが発現誘導されることを見いだした。神経細胞アポトーシスに於けるCAD、DNaseγの役割を明らかとするため、それぞれの活性化をCaspase非感受性ICAD変異体、及びDNaseγアンチセンスRNAを用い抑制した細胞株を樹立し、未分化神経細胞ではCADが、分化誘導に伴う自然発生アポトーシスではDNaseγがそれぞれDNA断片化を触媒することを明らかとした。
B細胞の成熟過程に於いては、自己反応性BCRを発現するB細胞がアポトーシスにより除去されることが知られている。我々はB細胞に於けるアポトーシスDNaseの発現を調べ、DNaseγが未成熟B細胞特異的に発現することを見いだした。未成熟B細胞株WEHI-231のアポトーシスに於けるCAD、DNaseγそれぞれの役割を上述の方法を用い解析し、薬剤誘導アポトーシスではCADが、BCR刺激誘導アポトーシスではDNaseγがDNA断片化を触媒することを明らかとした。
以上の研究成果により、分化過程にある細胞では未分化状態とは異なるアポトーシス経路により細胞死を起こすこと、当該経路に於いてDNaseγが中心的役割を果たすことが明らかとなった。分化過程にある細胞はAktなどの活性化により、一時的にアポトーシス耐性を獲得することが知られている。本研究により得られた知見は、DNaseγ経路がこのようなアポトーシス抑制のバイパスに重要な役割を果たす可能性を示唆している。
