Research Abstract

本研究においては、我々の胎児外性器、尾部形成過程の解析実績を基盤として尾部形成過程と協調して起こる内胚葉性器官(総排泄腔、尿生殖洞、VER (Ventral Ectodermal Ridge))発生過程を解析し、尾部形成における意義を考察した。
我々は遺伝子ノックアウトマウスを駆使することによって、胎児外生殖器、並びに尾部の形成に重要と推定されていたものの実体が分からなかったcloaca(総排泄腔)及びその周辺間葉において上皮間葉相互作用が存在すること、そして上皮側の主要な因子としてはShh、間葉側の制御因子群としては転写制御因子群が存在することを報告した。
さらにショウジョウバエなどを中心としてその付属肢先端における発生制御メカニズムに注目が集まっているDlx (Distaless)遺伝子群に注目し、Dlx遺伝子群の制御が総排泄空由来のソニックヘッジホッグ遺伝子下流にあることを初めて示した。このことは付属肢の発生過程において先端制御を担う制御因子が(外生殖器原器)発生場形成過程から上皮の重要な制御因子Shhの下流にあることを初めて示したものである。これらのデータは単なる外生殖器発生メカニズムの知見に留まらず胎児尾部形成、後部形成に重要な付属肢、器官形成のための遺伝子ネットワーク解明を初めて行ったものである。
