Research Abstract

アミロイド線維は、蛋白質が異常な立体構造をとり、線維を形成したものである。これまでに、アルツハイマー病βアミロイド線維(fAβ)、及びβ2-ミクログロブリン(β2-m)アミロイド線維等について、試験管内線維形成反応系を開発している。本研究ではこれらの反応系を用い、(1)種々の生体分子がアミロイド線維形成を促進・阻害する分子機構を、生体分子間相互作用の解析を通して疾病発症機構(医学)の観点から明らかにすること、及び、(2)種々の有機化合物による線維形成阻害・分解の分子機構の解明を目指して研究を行い、本年度は以下の成果を得た。
1.β2-mアミロイド線維の試験管内伸長において、生体に存在する伸長促進因子はまだ同定されていない。これを探索した結果、親水基に陰性荷電を持つリゾフォスファチジン酸など数種類のリゾリン脂質が線維伸長促進効果を有することを見出した。
2.試験管内でAβ蛋白質からのfAβ形成を阻害し、かつ形成されたfAβを不安定化する有機化合物を引き続き探索し、イブプロフェン等の非ステロイド性抗炎症剤、補酵素Q10及びドーパミン等の抗パーキンソン薬が強力な作用を示すことを見出した。
3.フラボノイドの示すfAβ線維形成・伸長阻害、不安定化の分子機構を解析した。ミリセチン(Myr)等の各種フラボノイドの、Aβ蛋白質モノマーとfAβに対する結合親和性を比較したところ、Myr等はfAβだけに高い親和性を示した。MyrはfAβに結合して特異的蛍光を生じるが、その結合形式は一次反応速度論に従うことを示した。以上より、MyrはAβ蛋白質モノマーの一次構造ではなく、fAβの高次構造を特異的に認識し可逆的に結合することで抗アミロイド効果を示すと考えられる。次いで、線維伸長活性に対する効果を検討し、線維断端に結合したMyrがAβ蛋白質の結合を阻害し線維伸長を阻害する可能性を示した。
