Research Abstract

抗体遺伝子のクラススイッチ(CSR)と体細胞突然変異(SHM)に必須の因子としてactivation-induced cytidine deaminase(AID)が単離されたが、いまだ分子レベルでの機序は明らかではない。SHMは抗体遺伝子の可変領域を標的としたDNA塩基置換であり、CSRは抗体遺伝子の定常領域のDNA配列を標的として、DNA2本鎖切断とその断端の結合による反応である。我々は、C末AID変異体により特異的にCSRだけが障害されることをすでに報告した。今回、研究代表者は、N末AID変異体がSHMだけを特異的に障害することを見つけた。これらの変異はAIDの活性中心ど考えられているcytidine deaminase motifには存在しないことから、N末あるいはC末にそれぞれ特異的に結合するcofactorが存在し、変異により各cofactorがAIDに結合できず、CSRあるいはSHMが障害されると予想される。つまり、可変領域を標的とするSHMにはN末に結合するcofactor、定常領域を標的とするCSRにはC末に結合するcofactorがそれぞれ必要で、cofactorを介してCSRとSHMという二つの反応が、標的遺伝子特異的に制御されているのではないかと推察された。
現在、C末AID変異体およびN末AID変異体のノックインマウスを作製中で、in vivoにおけるこれらのAID変異体の機能を解析するという課題が残った。
