Research Abstract

この研究では種多様化が急速に進んだと考えられる生物種群、特に東アフリカ・ビクトリア湖のシクリッド等を中心に、現存集団の遺伝的変異を解析することによって、集団のサイズや地理的構造などが、どのように種分化に寄与したかを明らかにすることを目的とする。ビクトリア湖のシクリッドについては岩場性、沿岸性の3属5種(Paralabidochromis)"rockkribensis",Lithochromis rubripinnis, L.rufus, Neochromis greenwoodi, N.rufocaudalis)でマイクロサテライト12遺伝子座の多型を調査した。沖合性二種(Haplochromis pyrrhocephalus, H.laparogramma)のデータも合わせて解析したところ、生態的に異なる岩場性・沿岸性・沖合性の種群間、及び種群内の種問に、弱いながらも階層的な遺伝的分化ができつつあることを見いだした。染色体の解析では東工大岡田グループと共同で単離したBACクローン24個のうち、20個が染色体マーカーとして使用可能なことが判明した。20個全てにおいて、マルチカラーFISH法による解析を終了させ、これら20個は18対の染色体を識別できることがわかった。一方TAARと呼ばれる化学受容体遺伝子ファミリーの進化について、9種の脊椎動物のゲノム情報を使って解析した。魚類では四足動物に較べ特に遺伝子数が増えており、また多くの遺伝子が嗅覚器官で発現していることが明らかになった。嗅覚遺伝子の分化が種の認識に関与している可能性もあり興味深い。更に、系統特異的に進化したシス転写制御領域の分子進化学を取り入れた比較ゲノムによる推定法と、トランスジェニックマウスを用いたシス転写制御領域の機能解析法の開発を行い、哺乳類特異的な転写制御領域を発見した。
