Research Abstract

アプリロニンAは、細胞骨格タンパク質のアクチンに作用する新しい型の強力な抗腫瘍性物質であるが、その生物活性研究を行うための試料量と誘導体が不足している。そこで、海洋動物の採集・抽出を行い、アプリロニンAを単離した。さらに、天然品の活性増強を目指して、強力はアクチン脱重合活性を有する海洋天然物ミカロライドBの部分構造を取り入れた人工類縁体の合成研究を行い、ラクトン部分の構成成分である重要合成中間体を合成した。
ハテルマライド・ビセライド類は、抗腫瘍性が期待されている海洋産マクロリドであるが、天然からの供給量がきわめて制限されている。そこで、既に申請者らが開発した合成経路を改良し、B-アルキル鈴木-宮浦カップリング反応を鍵段階とするハテルマライドNAの第2世代の全合成を達成した。この合成経路により、工程数の削減と通算収率の向上を達成できた。本合成経路は、類縁体であるビセライドの合成にも応用できるので、この合成を目指した研究も行い、ビセライドの重合合成中間体の合成も達成した。
オーリライドに関しては、アミノ酸をリジンに置き換えた誘導体やヒドロキシル基を官能基化した誘導体などいくつかの人工類縁体を合成し、その構造活性相関研究を行った。その結果、標的分子探索に用いるプローブ分子合成に用いることができる官能基を備え、細胞毒性を保持した人工類縁体を創成することができた。今後は、この分子を用いたプローブ分子の合成と評定分子の探索を行う。
