Research Abstract

細胞情報伝達系に特異的に作用する活性天然物質(生体機能分子)を発見して広く世に供給することができれば、その活性物質がツール(生化学試薬)として利用されるとともに、新たな医薬品開発に展開することが期待される。本研究では、1)特にがんと神経変性疾患に着目し、これら疾患に関わる細胞内情報伝達系に特異的に作用する活性天然物質を探索するアッセイ系を構築し、これまでに集めた海綿を中心とする海洋生物の抽出エキスライブラリーから探索する。2)そして新たに発見した阻害物質とそれらから設計・合成した光親和性放射性標識体やビオチン標識体などの各種の天然分子プローブを用いて標的分子を同定し、細胞内情報伝達系への作用機構を解析する。本年度は、以下の成果を得た。1)ヒト月臍帯静脈内皮細胞HUVECに対して3000倍以上の選択性を持って増殖抑制活性を示す活性成分cortistatin類を見出した海綿の抽出エキスを精査し、合計11個の類縁体を見出し、それらの化学構造を明らかにするとともに、構造と活性の相関を調べた。2)Cortistatin Aの簡単なモデル化合物では血管新生阻害活性が発現しないことが判明したため、その全合成研究に着手し、CD環セグメントの合成に成功した。3)p21プロモーター領域を活性化する物質を探索する活性試験法を用いて、アユルベーダ薬草からcryptolepineを、海洋微生物の培養物エキスからsecalonic acid Dを活性物質として見出し、それらの作用を詳しく解析した。4)潜伏期間の結核菌に対しても生育阻害活性を示す抗菌物質を探索し、海綿から強力な活性物質halicyclamine Aを見出し、その作用機構について解析した。
