Research Abstract

一般に植物は動かないものと考えられているが、マメ科植物は、昼間は葉を開き、夜になると葉をたたんで「眠る」、就眠運動と呼ばれる特異な生物現象を行う。これは、紀元前、アレキサンダー大王の時代から知られている、非常に興味深い現象である。本研究では、これらの興味深い生物現象を、分子レベルでの解明を試みた。
我々は、マメ科植物の就眠運動は、就眠・覚醒両物質によってコントロールされており、それらが属特異的であることを見いだしている。そこで、Albizzia属アメリカネムノキの就眠・覚醒物質に蛍光色素を結合させた蛍光プローブを合成し、2つの活性物質の作用部位を調べた結果、就眠・覚醒両物質は、葉枕部内側の運動細胞に偏在する受容体に結合することで、葉枕の片側の運動細胞のみに体積変化を引き起こし、これを膨潤・収縮させることがわかった。さらに、就眠物質の両エナンチオマーの蛍光プローブを用いた蛍光標識実験「エナンチオ・ディファレンシヤル分子プローブ法」により、葉枕内側の運動細胞に、就眠物質の立体化学を認識する受容体が含まれることを強く示唆する結果を得た。一方、Lespedeza属メドハギの運動のリズムをつくりだす鍵酵素β-グルコシダーゼの精製に成功し、これが分子量150kDaの巨大な糖タンパク質であることを明らかにした。
