Research Abstract

真核細胞において生命現象の中軸を担う細胞周期、血管新生、細胞死の素過程には未解明な部分が多く、これらを制御する新規天然有機化合物の探索・創製・応用研究の意義は大きい。本年度は、主としてこれまでに確立した評価系を利用して、微生物代謝産物をスクリーニング源として探索研究を行うとともに、複数の生理活性小分子に関する化学生物学的な解析研究を行った。
マウス乳がん細胞FM3Aのcdc2キナーゼが温度感受性になった変異株tsFT210細胞においてG2/M期停止作用を有する放線菌ストレプトミセス属A2215株の培養抽出物を見出した。そこで、培養上清をn-BuOH抽出後、Diaion HP20、ODSカラムクロマトグラフィーにより、順次、活性物質の単離・精製を行い、活性物質として淡黄色不定形固体RKGS-2215Aを得た。FAB-MSおよび各種NMRスペクトルの解析結果より、RKGS-2215A(C31H48010)は新規ポリエン化合物であることを明らかにした。フローサイトメトリー解析、ならびに細胞染色解析の結果、RKGS-2215Aは1 μg/m1の濃度でtsFT210細胞の細胞周期をM期に停止することが明らかになった。
また、我々がこれまでに見出している糸状菌由来の血管新生抑制剤エポキシキノールBの詳細な作用機構解析研究を行った。その結果、エポキシキノールBは、ヒト臍帯静脈血管内皮細胞(HUVECs)において、VEGFR2に不可逆的に結合し、VEGFによって活性化される細胞内情報伝達系を抑制することを明らかにした。
