Research Abstract

研究目的
皮膚幹細胞の培養法の確立によりアトピー性皮膚炎患者の組織及び培養皮膚組織モデルにおける微量金属元素のイメージングと化学状態分析を非破壊的に単一細胞レベルで行い、その後同一試料に対して組織学的、遺伝学的検討を行うことによって、アトピー性皮膚炎における病態(組織の剥落、バリア機能の阻害、サイトカイン等)の制御と微量金属との相関、そして制御に働く候補遺伝子を明らかにすることである。
研究経過
Spring-8にて蛍光X線解析による元素分布イメージングと細胞内微量元素のXAS分析により化学状態の解明を行った。
研究結果概要
(1)アトピー性皮膚炎患者3名からのサンプルをとり、培養を行った上で、固定サンプルを作成した。これらのサンプルを用いて、放射光装置で分析を行い、金属分布についてコントロール細胞との比較を行った。
(2)アトピー性皮膚炎患者のサンプルを培養し、サンプルをLPSなどの刺激前後でcDNAを採取した。一方、候補遺伝子については、DNAチップを作成し、これを用いて、患者サンプルにおいて、皮膚細胞で刺激前後で発現が変化する遺伝子を特定するシステムを作成した。
