Research Abstract

本研究では主に遺伝子ノックアウトマウスの解析を通して、プロテインチロシンホスファターゼζ(PTPζ)とNa_xイオンチャンネルの関わる生命現象とその分子・細胞機構を明らかにすることを目指す。本年度の研究成果は以下の通りである。
a)PTPζのシグナル伝達系
1.基質分子中の基質サイトの同定
PTPζの基質分子中の基質サイトの同定については、昨年度p190 RhoGAP中のY1105が明らかになったが、本年度はGit1(ArfGAP)とMagi1について、そのサイトを決定した。
2.PTPζ遺伝子ノックアウトマウスのメタンアンフェタミンに対する不感受性の分子機序について、ドーパミン・トランスポーターの細胞内への取り込み能について細胞生物学的解析を行なった。
3.眼におけるPTPζのisoformの多様性について解析を行なった結果、受容体型PTPζがsheddingを受けている可能性が明らかになった。
b)Na_xチャンネル
1.昨年度Na_xとNa^+,K^+-ATPaseのα1サブユニットが結合していることを見出したが、その後この結合が細胞内Naレベルの上昇に応答してNa^+,K^+-ATPaseが活性化するために必要であることが明らかになった。また、Na_x陽性細胞ではNa^+,K^+-ATPase活性化の結果、糖代謝の増加、並びに乳酸の産生と分泌が上昇することが確認された。
2.マウスの尿中におけるバソプレッシン量を測定する系を確立した。
