Research Abstract

The effects of glutamate on postsynaptic neurons are mediated by iGluRs in the dendritic plasma membrane. The iGluRs are accumulated at postsynaptic specializations, and a modification of synaptic efficacy by changing number and/or density of AMPA type iGluR at the postsynaptic specialization may be relevant to learning and memory. To study LTP-induced changes in distributions of AMPA and NMDA type iGluRs in the postsynaptic specialization of hippocampal neurons, we employed SDS-digested FRIL, with which 2-dimensional distribution of the membrane proteins can be analyzed quantitatively at &lt ; 20 nm resolution, and revealed that AMPA receptors, but not NMDA receptors, are abundant on the dendritic plasma membrane and AMPA receptors at the postsynaptic specialization on the dendritic spine, but not on the dendritic shaft, are regulated more dynamically than NMDA receptors after LTP induction.
