Research Abstract |
In this study, we found that an acute exercise induces an increase in plasma LPS concentration. Furthermore, an increase in OVA specific IgE concentration in plasma were greatly inhibited by LPS injection. Th1/Th2 balance was also changed Th1 domination by LPS/OVA stimulation. In addition, a production of IL-12 which is a cytokine for inhibition of allergic reaction, from peritoneal macrophages in tlr-4 gene mutated mice was significantly lower than that in wild type mice. Accordingly, we thought that exercise-induced increase of LPS might prevent reaction of OVA induced allergic reaction. Moreover, we investigated the effect of LPS on OVA-induced anaphylaxis as one of allergic reactions. Release of histamine, which is one of mediators in anaphylaxis, was also examined. Additionally, we tried to examine the possibility of macrophages-dependent anaphylaxis by using the technique of macrophage deletion. As the results, we observed the inhibition of OVA specific IgE and IgG1, and then plasma histamine concentration was also inhibited in spite of OVA re-exposure. However, OVA re-exposure induced reduction of rectal temperature as an index of OVA-induced anaphylaxis was not inhibited by LPS stimulation. Also OVA-induced anaphylaxis in macrophage deleted mice was not inhibited. These result suggested that LPS might not prevent allergic reaction although LPS in which exercise induces an increase, inhibits both of antibody production and histamine release.
