Research Abstract

The purpose of this study is to find the genes that enhance the resistance against oxidative stresses. To this end, we searched genes that highly expressed in hydrogen peroxide (H_2O_2) resistant cells, named as HP50 and HP100 cells. We found 178 genes and 240 genes that expressed more than three-times in HP50 and HP100 cells as compared with parent cell HL-60 cells, respectively. Among these genes, we focused on the 85 genes and further confirmed by Real-time PCR. In HP50 and/or HP100 cells, 44 genes were expressed more than twice as compared with HL-60 cells. And 15 of these 44 genes were induced under the oxidative stress (H_2O_2, AAPH and/or DEM) in HeLa, HepG2 and/or HL-60 cells.
At the moment, we are constructing the expression vectors to evaluate the function that really contribute to obtain the resistance against oxidative stresses. Some of these genes enhanced resistance against oxidative stress when these genes were overexpressed in the HeLa cells, but the effect was not remarkable. Then we are preparing the stably expressed cells and try to make clear the function of these candidates.
