Research Abstract

Petunia Lateral-shoot Inducing Factor (LIF) is specifically expressed at the basal region of axillary buds and drastically increases lateral shoot branching when overexpressed in transgenic plants (LIF-OX). In this study, we investigated the molecular function of LIF in the control of lateral shoot branching with a focus on its relation to phytohormones. We also performed a comprehensive functional analysis of SUP/LIF family genes in Arabidopsis (Superman-Like, AtSPL) and found that AtSPL5, a new AtSPL gene, is implicated in responses to environmental stresses and phytohormones.
Determination of phytohormone contents indicated that the levels of cytokinin metabolites in LIF-OX petunia plants were significantly changed in comparison to wild type plants, with remarkable decrease in basal t-Zeatin level, which we presume to correlate with the promotion of lateral-shoot branching by LIF. Microarray analysis using LIF-inducible transgenic Arabidopsis plants in the GVG system revealed a number of LIF-regulated genes including those for transcription factors and tau-type GSTs.
The expression of AtSPLS in young seedlings was downregulated in response to environmental stresses, drought, salt, ozone and UV-B ; and phytohormones salicylic acid (SA) and abscisic acid (ABA). RNAi-mediated knockdown of AtSPL5 resulted in an increase in expression of ABA-responsive genes rd29A/B. Given that AtSPLS contain an EAR-motif-like transcriptional repression domain at its C-terminal, our results suggest that AtSPL5 is involved in suppressing the expression of stress responsive genes of plants during their growth under normal environmental conditions.
