Research Abstract |
Using human B-cell lines in which vIRF1 was expressed in a doxycycline-inducible manner, it was demonstrated that vIRF1 expression downregulates p53 protein level at the posttranslational level. It also was demonstrated that vIRF1 expression enhanced p53 cytoplasmic localization, resulting in an increase of mdm2-mediated p53 ubiquitination and degradation. Furthermore, vIRF1 interacts with ATM kinase. The interaction leads to the inhibition of etoposide-induced ATM activation and p53 phosphorylation. In summary, vIRF1 interacts with ATM and inhibits ATM/p53 signaling pathway.
