Research Abstract

The patients is a 23-year old women who has a family history of Familial Mediterranean Fever (FMF) and since the age of 11 years has had recurrent chest and abdominal pain with fever. Genomic DNA analysis of the patient revealed the E148Q/M6941 mutation. Her mother also has a heterozygous M6941 mutation and experienced the same symptoms until 30 years of age. To establish the patients (mutated) cell line, we tried to transfect with pSV40 ori (plasmid DNA of SV40 virus deleted with origin) using lipofectin method. It was unable to establish the cell line. So we compared the levels of interokin-1 beta (IL-1beta) between the sample from each patient (E148Q/M694I mutation and M6941 mutation) and healthy control. Cells were treated with LPS for 48 hr. Supernatants were collected and assayed for IL-1beta by ELISA. IL-1beta ELISA was performed according to the manufacture's instruction. There was no statistically significant difference in the levels of IL-1beta among three samples. We also compared the levels of IL-1beta in the presence or absence of colchicines from three samples. There was no statistically significant difference in the levels of IL-1beta between any of the samples.
