Research Abstract

The purpose of the study is to examine the function of the brain including thalamus and hypothalamus, by the pupil fluctuation.
It was shown that a quantity of pupil fluctuation increased by stress sleepless. The cycle of pupil fluctuation of the time was about 1Hz. On the other hand, in a physically unimpaired person, the emotional loading test did not cause the meaningful change of pupil fluctuation.
Concerning the awaking process from anesthesia, the difference about the pupil fluctuation was not seen between the intravenous anesthesia (propofol) and the volatile anesthesia (sevoflurane), both of which are GABA agonist. However, in the case of ketamine which was NMDA antagonist, the lability of eye movement itself was recognized in an awakening process. A consciousness state is controlled by processing source of information and nerve modification action in addition to an activation level of a brain. The source of information means the relative balance between outside source and internal source, and can be recognized with movement of the eye, which is a substitute for the PGO-wave. Therefore, the result may suggest that NMDA-antagonist affects the gating of input/output system in the brain.
Furthermore, in the spectral analysis simultaneously performed, ketamine, an NMDA-antagonist, increased the spindle peak of the α-band (which was usually induced by GABA-agonist) to 4 Hz higher. In the phase analysis to examine the phase coupling at that time, the peak of the bicoherence also shifted to 3-4Hz higher on the frequency domain.
The result suggest that ketamine work the reticular thalamic network as constituting the consciousness. The result showed that the measurements of the fluctuation of pupil and eye movement are important to examine the awaking consciousness, together with electroencephalogram.
