Research Abstract |
Stress is indicated as a potential risk factor for periodontitis but the detail mechanism of both association is still unclear. We previously showed that neuropeptide is involved in bone resorption using rat experimental periodontitis model. There are several neuropeptides which are supposed to act in the local periodontal tissue so we investigated those gene expression comprehensively using microarray. A nylon ligature was placed around the second right maxillary molars of wistar rats. The animals were then divided into group S, exposed to a restraint stress for 12 hours/day for up to 10 days, and group N, controls. Three animals were sacrificed on days 2, 4, 6, 8 and 10. The total RNA was isolated from gingiva around the second right maxillary molars and then synthesized fluorescence labeled cRNA. The gene expressions in gingiva were analyzed using microarray which competitive hybridization method is utilized. 6 genes which is involved stress response in general were found using gene ontology (GO SLIMS) functionally classified from 83 genes which those expression increased twice in stress group. The gene expression of Tachykinin-1, which is represented by substance P and neurokinin, increased in day 2 in group S. At days 4, several chemokines, interleukin-1β, interleukin-6, were upregulated. We will analyze more detail of those gene expression and it might be possible for adopting siRNA which inhibit those gene expression to reduce the influence for periodontal tissue.
