Research Abstract

細胞内のCaをセカンドメッセンジャーとするCaシグナリングは、数多くの受容体の下流に存在する重要な信号伝達機構の一つである。
T型リンパ球や脳にも存在するADPリボシルシクラーゼ活性の大部分はCD38抗原であることがわかってきた。しかし、脳内にはCD38以外の新しいADPリボシルシクラーゼ活性を持つ分子が存在することを考え、脳型ADPリボシルシクラーゼの精製を行うため、野性株マウス脳内のシクラーゼ活性を比較した。
(1)ジクラーゼ活性は、驚いた事に視床下部に高く、続いて小脳や下垂体後葉であった。
(2)CD38のメッセンジャーRNA発現を調査したところ、活性値の分布と同じであった。
(3)CD38ノックアウトマウスでは、シクラーゼ活性は極端に低く、視床下部でもほとんど活性やmRNAは検出できなかった。
(4)シクラーゼの酵素活性産物であるサイクリックADPリボース(cADPR)濃度は、野生型マウスに比して、CD38ノックアウトマウスでは低かった。
(5)視床下部に細胞外からcADPRを与えたところ、オキシトシンの分泌は見られたが、ノックアウトの視床下部からは検出できなかった。この事から、CD38がcADPRのトランスポート活性を有している事も判明した。(Nature 446,41-45,2007)
以上、脳型シクラーゼの90%がCD38である事を種々の方法で証明した。当初の目的のCD38以外のシクラーゼの精製は、非常に困難な課題である事が判明した。
