Research Abstract

多孔質円筒体を以下のように作製した。ポリウレタンをテトラヒドロフランに溶解させた後、塩化ナトリウムを加え、粘調な溶液を調整した。この溶液にガラス棒を浸漬後、ゆっくりと引き上げ、室温で乾燥させた。これを数回繰り返し、所定の外径に調整した。完全に乾燥後、水中に数回浸漬させて塩化ナトリウムと残留溶媒を除去した。
作製した多孔質円筒体をガス滅菌し、培養液中に浸漬した状態でヒト羊膜上皮細胞を内径面に播いた。ヒト羊膜上皮細胞が生着し、内腔面が上皮細胞に被覆されていることを確認し、両端にラットから摘出した気管を端々吻合させてラット気管吻合多孔質円筒体を形成した。形成したラット気管吻合多孔質円筒体をラット皮下に移植し、約30日生体内培養を行った。その結果、作製した多孔質円筒体は生体親和性があり、羊膜上皮細胞が内腔に良く生着することが確認できた。生体内培養実験では、ヒト羊膜上皮細胞が多孔質円筒体の内腔に生着していると、ヒト羊膜上皮細胞に置き換わるようにラット由来気管上皮細胞が浸潤してくることが示唆された。さらに、ヒト羊膜上皮細胞が多孔質円筒体の内腔に生着していると、外径表層から浸潤してくる繊維芽細胞の内径空間部への侵出が抑制されることが示唆された。
