Research Abstract

β1,3グルカンが作る2重螺旋には、ピッチ1.8nmの61螺旋の溝ができる。この溝の大きさは核酸塩基を取り込むのに最適の大きさがあり、溝の奥には水素結合サイトが存在することが分かった。事実、コンピューター上で、核酸塩基をその溝に入れてみると極めて容易に包摂される。従って、核酸塩基の親水と疎水性のバランスを化学的に調整さえすれば、GMP、GDPが再構成した多糖の3重螺旋内に内包されると考える。しかし、現在、かなり苦戦をしている。そこで、今年度は、アプローチを変えて、最も疎水性の高いと考えられる、カーボンナノナノチューブの内包に挑戦した。β1,3グルカンがカーボンナノチューブ、特に、単層ナノチューブの優れた可溶化剤になることを見出した。興味深いことに、グルカンがナノチューブ表面をらせん状に被覆し、グルカン特有の周期構造をナノチューブ表面でも再生していることがAFMによる観察の結果から明らかとなった。カーボンナノチューブ表面に規則的なパターンを形成することはナノチューブへの機能付与を考える上でも極めて重要であることは言うまでもない。今後、本系がナノチューブの機能化とバイオテクノロジーへの展開において、重要な役割を果たすものと期待される
