Research Abstract

作物の根は分泌層で覆われており、この層を介して根圏土壌中の物質が吸収される。本研究は、根-土壌界面に存在する分泌層内での物質の動きを明らかにする方法論を検討し、安定同位体でラベルした水や窒素が根圏から分泌層を介して根面へ取り込まれる過程を明らかにすることを目的として、以下の研究を実施した。今年度は、昨年検討したシリコン板法とともに、寒天培地法も含めて検討を継続するとともに、飛行時間型2次イオン質量分析計(Tof-SIMS)による安定同位体の取り込みに関する実験も実施した。まずシリコン板法では直接、粘液を観察することが困難であった。そこで様々な濃度の寒天培地を試験したが、同様に寒天への封入では、真空中での脱水により寒天がくずれてしまいうまく観察ができなかった。そこで、やはりクライオシステムで観察する必要があることを認識し、まずは植物サンプルでの飛行時間型2次イオン質量分析計での観察手法の確立を先決課題として実施した。イネでは真空中での組織の保持がうまくできなかったため、茎や'根の直径の大きなダイズを用いて、真空中での観察を実施した。クライオミクロトームで切断した根と茎切片を観察したところ、すべてのイオンの総和であるトータルイオン観察によって木部導管や師部組織を観察することが可能となった。次に安定同位体として簡易な暴露が可能な重水を用いて、一定時間吸収させた後の重水の分布を観察したところ、木部導管部位では重水の組織への吸収を認めず、この手法での同位体の吸収を観察することが可能であることを確かめた。次にクライオシステムで同様の観察を行ったところ、より鮮明な観察が可能となった。今後はこれまで試したシリコン板法や寒天培地法をクライオシステムで観察する実験系を確立する必要がある。
