Research Abstract

生体の3次元断層画像を得る方法(in vivoイメージング)の一つであるMRI(Magnetic Resonance Imaging)は、放射線被曝がなく、かつ生体を傷つけることなく深部にわたる画像を得ることが出来るという大きな利点を持っている。そのためMRIは画像診断法として汎用されており、その画像は水分子の水素原子核におけるNMR現象を利用している。前年度までに生体内でのZn(II)の動態をMR画像変化としてバイオイメージングできるMRI造影剤の開発を行った。
今年度は昨年度後半から行っている遺伝子発現をモニターできるベータガラクトシダーゼプローブの実用化を目指した。Gd(III)は強い常磁性イオンであるため、通常、イオン近傍の水素原子核の主に縦緩和時間を短縮する。Gd(III)に配位している水分子が強いMRシグナルを示し、水分子の配位数の変化はこのMRシグナルを変化させる。今回、このような原理とは異なり、アルブミンのような高分子量の分子とGd(III)錯体が結合することでGd(III)錯体分子自身の分子回転が非常に遅くなり、周囲の水分子に対して、大きな緩和時間の短縮を引き起こす。この現象はreceptor induced magnetization enhancement(RIME)と呼ばれるが、これを利用した分子設計を行った結果、アルブミンとGd(III)錯体との結合が酵素により切断されることでMR信号強度に大きな変化を生じさせることに成功した。更に、アルブミンとの結合の親和性をより高める検討を引き続き行っている。
