Research Abstract

pHtrII(pharaonis halobacterial transducer protein)は2回膜貫通型の膜蛋白であり,膜中でpharaonis phoborhodopsin(ppR)と2:2のモル比で複合体を作り,光信号を細胞内へ伝達する.pHtrIIは大腸菌やサルモネラ菌のアミノ酸への走化性のレセプターと類似しており,膜ヘリックス部分,膜から出た直後の部分(linker region),メチル化サイトおよびリン酸化サイトからなる.大腸菌の走化性の実験からは,linker regionは重要な役割をしていると考えられている.そこで,ppR/pHtrII系でもliker regionが信号伝達に重要ではないかとの考えのもとに実験を進めた.従来,ppR/pHtrIIの結合は膜内ヘリックス間相互作用で結合される考えれていた.linker regionのみを大腸菌で発現させ,liner regionのみでもpHtrIIに10μMの結合定数で結合することを等温滴定型熱量計で示した.さらに,linker regionを15Nでラベルしたサンプルを用い,NMRの信号のアサインをし,ppRの存在によって,Chemical shiftが変化することを示した.また,このlinker regionのCDは界面活性剤の有無により変化するので,多くのコンフォメーションをとることの出来る「カメレオン」蛋白であることが分かった.このような性質が信号伝達に重要な役目をしているのであろう.
