Research Abstract

本研究は,ドラッグをデンドリマーの手法を用いて球の表面に一定方向で固定したナノアセンブリー分子を設計・合成し,そのドラッグが本来持つ生物活性がどのように変化するか,キラリティーを超えるドラッグを作り出すことができるか,の2点を明らかにすることを目的とする.この目的を達成するためのリード化合物としてブレフェルジンA,スワインソニン,免疫抑制剤FTY720を選択した.本年度(最終年度)は,前年度までに分子設計・合成した種々のナノアセンブリー分子の鶏胚漿尿膜(CAM)法による血管新生阻害活性について評価した.免疫抑制剤FTY720は,冬虫夏草から単離された免疫抑制作用を示すISP-1(myriocin)の化学修飾によって不斉炭素を除去した,つまりキラリティーをなくした合成化合物であり本研究の目標に適したリード化合物である.FTY720のアルキル側鎖をアセチレンに置換したTX-2148(モノイン),TX-2152(ジイン),TX-2256(トリイン)の血管新生阻害活性はそれぞれ42%,90%,60%(10μg/CAM)であり,TX-2152はFTY720(77%)と同等以上の阻害活性を示した.この結果より,FTY720のオクチル基に対するバイオアイソスターとして疎水性及び分子サイズの大きく異なるジイン(もしくはトリイン)構造が示された.その理由として,剛直なポリイン構造によって分子の自由度が制限されるとともに,標的分子との相互作用の際の方向性が規定されたためではないかと考えられる.以上より,一定方向で固定したナノアセンブリー分子の生物活性がプラスに変化する事例を見出すことに成功したといえる.
