Research Abstract

1)ヒトemmprinの第一Igドメイン由来のペプチドを20アミノ酸ずつ4本に分け、それぞれemp#1-4とし、cocultureにおけるMMP-2産生亢進への阻害作用を検討すると、emp#2のみに阻害活性が認められ、それは脳腫瘍細胞株、ATL細胞、悪性黒色腫、肉腫細胞株(epithelioid sarcoma(ES),malignant peripheral nerve sheath tumor(MPNST))と線維芽細胞のcocultureにおいて共通して認められた。この阻害活性はemp#2を10アミノ酸ずつに2分すると失われた。
2)上記のemp#2ペプチドによる阻害活性は、emp#2と腫瘍細胞とのpreincubationにより認められ、emp#2と線維芽細胞とのpreincubationでは認められなかった。この結果より、emp#2による阻害は当初考えられていたcompetitive inhibitionによるdecoy moleculeとしての作用では無く、emmprinの腫瘍細胞膜上でのhomodimerまたはheterodimer(multimer)形成によると考えられた。Polylinkerを用いたdimerization studyでは、emp#2によってemmprin相互のhomodimerizationは抑制されなかったが、heterodimer(multimer)形成阻害の予備実験では阻害を認め、現在確認中である。
3)Full emmprin cDNA, N-terminal tagged emmprin cDNA, C-terminal tagged emmprin cDNA-expression vectorを作成し、tissue inhibitor of matrix metalloproteinases(TIMP)のsignal sequenceを用いたemmprin活性阻害ペプチド(emp#2)分泌plasmidをoverlap extension PCR法にて作成し、その阻害活性の確認中である。
