内在性分泌型RAGEタンパクの病原微生物に対する防御作用

Research Abstract

本研究の目的は、「内在性分泌型RAGE(esRAGE)タンパクは、自然免疫・粘膜防御を担う重要な液性因子である」という仮説を検証することである。
1.esRAGEと病原微生物、特にListeria monocytogenesとの相互作用の解析
(1)表面プラスモン共鳴法を用いた実験において、esRAGEとL.monocytogenesが直接結合することが明らかになり,そのリガンドは鞭毛タンパクflagellinであった。その結合解離定数は約10^<-8>Mであった。
(2)esRAGEタンパクが鞭毛タンパクflagellinと結合することで、L.monocytogenesの運動を抑制した。増殖阻害活性、殺菌作用、オプソニン効果は認めなかった。
(3)flagellinはesRAGBのみならず膜型RAGEにも結合することがわかり、その結合によってRAGE細胞内シグナルであるNFκBが活性化が生じた。
2.RAGE/esRAGEを共に欠損するRAGEノックアウトマウスを用いた感染実験
(1)RAGE+/+マウス由来腹腔マクロファージの方が、RAGE-/-マウス由来腹腔マクロファージよりL.monocytogenesの処理能力が高かった。
(2)RAGE/esRAGE系はマウスのリステリア感染に対して防御的に働いていた。
a.2種類の遺伝背景のマウスで感染実験を行った。B6、CD-1ともにRAGE+/+マウスの方がRAGE-/-マウスに比べて有意に生存率が高かった。
b.定量的に臓器単位面積当たりのL.monocytogenesの病巣部数を数えたところ、RAGE-/-マウスの方が有意に多かった。
c.組織学的炎症の程度もRAGE-/-マウスの方が有意に重症であった。
以上の結果から,esRAGEのみならず膜型RAGEを含めたRAGE/esRAGE系は,病原微生物に対する防御作用を担う生体因子であると考えられた。
