Research Abstract

我々がクローニングしたJNK特異的dual specific phosphatase (DUP)をコードするDUP16遺伝子とそのシステインをセリン置き換えたdominant negative DUP16遺伝子をT細胞にのみに発現する1ck-promoterで発現させたトランスジェニック(Tg)マウスをもちいて、Th1/Th2応答を検討した。TgマウスのナイーブT細胞を用いて抗CD3抗体/抗CD28抗体で刺激しIFN-γ、IL-4、IL-2の産生を調べたところ、DUP16-TgマウスでIL-2,IL-4産生の低下が、dominant negative抑制された。Mycobacterium bovis (BCG)と卵白アルブミン(OVA)に対するTh1/Th2応答を検討した。BCGを感染させて、臓器内菌数を調べたところ、DUP16-Tgマウスで菌数の低下、dominant negative DUP16-Tgマウスで菌数の増加を認めた。さらにPPD抗原で刺激した細胞内サイトカインによってTh1機能分化を調べたところ、DUP16-TgマウスでTh1細胞数の増加、dominant negative DUP16-TgマウスでTh1細胞数の低下を認めた。これらの結果よりT細胞に発現されるDUP16はTh1細胞の産生・維持にはネガティブに働く可能性が示唆された。次にこれらのTgマウスに卵白アルブミン(OVA)/CFAで免疫して、OVA特異的抗体産生を調べたところ、DUP16-TgマウスIgEの産生が低下、dominant negative DUP16-Tgマウスで上昇した。OVA特異的CD4Th細胞のサイトカイン産生を細胞内サイトカインによって調べたところ、DUP16TgマウスでTh2細胞応答の有意な低下を,dominant negative DUP16-Tgマウスで上昇を認めた。これらの結果よりT細胞に発現されるDUP16はTh2細胞応答を制御することによってTh1/Th2応答を制御すると考えられる。
