Research Abstract

日本人の平均寿命は、世界の中でトップとなるに至ったが、その一方で急速な高齢化が大きな社会問題となり、老化の予防法の確立が急務となっている。酸化ストレスが老化の促進に重要な役割を果たしていることは多くの研究が示している。最近、テロメア繰り返し配列(5'-TTAGGG-3')nの短縮が、酸化ストレスにより促進するとの報告がなされた。本研究では、酸化ストレスによるテロメア領域の短縮促進機構を明らかにし、さらに、老化促進モデルマウスを用い、最新のプロテオミクス解析を駆使して老化の分子マーカーを同定することを目的とした。その結果、銅存在下で過酸化水素により5'-GGG-3'配列中の5'側Gが酸化されることが明らかになった。また、一酸化窒素と活性酸素共存下においても5'-GGG-3'配列中の5'側Gが著しく損傷された。さらに、正常線維芽細胞WI-38においてUVA照射量に依存して、酸化的DNA損傷の指標である8-oxodG量の増加とテロメア配列の短縮促進が認められた。以上の結果から、テロメア領域の短縮促進には、酸化ストレスによる塩基配列特異的DNA損傷が重要な役割を果たしていることが示唆された。
高酸化ストレス状態にある老化促進モデルマウス(SAM)を用いてプロテオミクス解析を行った結果、Cu, Zn-SODやselenium binding protein等の抗酸化タンパク質および78kDa glucose-regulated protein等の分子シャペロンが酸化傷害を受けている事が明らかになった。これらの酸化障害に重金属と過酸化水素が重要な役割を果たしていることが示唆された。さらに、methionine adenosyltransferaseやmajor urinary proteinは老化のバイオマーカーとなる可能性が示唆された。
これらのことから、抗酸化剤が老化防止に有効である可能性が示された。
