Research Abstract

従来、多くの発癌予防試験はPKC活性化剤であるTPAを用いた発癌誘導に対する予防効果の検討によってなされてきたが、TPAによる発癌誘導機構は不明であった。我々はサイクリン依存性キナーゼの阻害因子として細胞増殖抑制に働くp18遺伝子の発現低下が、TPAによる発癌誘導に深く関与していることを見出した。
本研究ではこのp18遺伝子発現低下を回復させる新規発癌抑制物質のスクリーニング系構築を目指し、まず既に発癌予防効果が指摘されている食品成分について試行したところ、TPAによるp18発現低下を、ゲニステインとクルクミンが回復させうることを見出した。特にゲニステインはチロシンキナーゼ阻害活性を有することが知られており、チロシンキナーゼの関与が考えられた。ハービマイシンA等のチロシンキナーゼ特異的阻害剤を用いても、TPAによるp18発現低下が回復されたことから、この経路に実際にチロシンキナーゼが関与している可能性が考えられ、現在その同定を行っている。
一方で、ケルセチンがTPAによるp18発現低下を促進してしまうこともわかった。ケルセチンは発癌予防効果だけでなく、場合によっては発癌促進に働くとする報告も散見され、今回の結果でその分子機構が説明出来るかも知れない。
本研究でゲニステインやクルクミンのような発癌抑制物質と、またケルセチンのような発癌促進に働く可能性のある物質を見出せたことで、TPAによるp18発現低下に着目したスクリーニング系の有用性が示された。同時に、発癌プロモーターを用いた動物発癌実験系が、ヒト発癌において中心的な役割を果たすRB遺伝子の失活に至る経路と密接に関連していることと、癌予防におけるRB経路の深い理解が極めて重要であることがあらためて示された。
