Research Abstract

ミトコンドリアはDNA修復酵素系に富み,ダイナミックにミトコンドリアDNA (mtDNA)変異の修復を受けていることがわかってきた.mtDNA修復系の特徴から,Chimeric RNA/DNA oligonucleotidesの有効性が期待されている.海外共同研究者のd'Alarcao教授らはoligonucleotidesをアルキルトリフェニルホスホネート(APP)誘導体化すると,ミトコンドリア内へ効率よく集積することを発見した.本研究では,mtDNA変異を標的としたChimeric RNA/DNA oligonucleotidesを作成し,APPで誘導体化して,ミトコンドリア集積性を高めて,mtDNA変異を標的とする一塩基置換修復法を開発する.
1.ミトコンドリアDNAを分子標的とする特異的一塩基置換修復法の開発
ミトコンドリアDNA3243A/G変異を標的とするChimeric RNA/DNA oligonucleotidesを化学合成した.正常ミトコンドリアDNAと3243G変異DNAのmixtureをテンプレートとしたミトコンドリアDNA複製系に,Chimeric RNA/DNA oligonucleotidesを添加すると,容量依存性に3243G変異が修復されることが明らかにした.
2.化学合成Chimeric RNA/DNA oligonucleotidesのアルキルトリフェニルホスホネート(APP)誘導体化
Chimeric RNA/DNA oligonucleotidesを化学合成し,アルキルトリフェニルホスホネート誘導体(APP)による修飾が可能か,検討した.対照としたDNA oligonucleotidesに比べて,Chimeric RNA/DNA oligonucleotidesは,APP化修飾効率が対照の約20%と低いことが明らかになった.
