川崎病との関連が推定される新規ウイルスの同定と川崎病の病因解析

Research Abstract

平成16、17年度までにTNF-α刺激HUVECおよび正常心筋由来cDNAライブラリーと回復期患児血清、あるいは川崎病急性期患児抹消血単核球とγ-グロブリン治療歴のない急性期患児血清とでSEREX解析を行ない、それぞれ69個、19個、8個の陽性クローンを単離した。平成18年度はそれぞれがコードする遺伝子/抗原の川崎病との関連について検討した。HUVECから最も高頻度で単離されたトロポミオシン(18/69)について組換えタンパクを用いて27名の川崎病患児血清と9名の非川崎病患児血清についてのELISA解析を行なった。川崎病患児血清中抗トロポミオシン抗体は非川崎病患児血清に比べ有意に高い値であった。さらに急性期川崎病患児と回復期川崎病患児を比較すると急性期川崎病患児血清中の方が有意に高い値を示した。このことはトロポミオシンの川崎病発症あるいは病態への何らかの関与を示唆している。正常心筋より同定されたうちの3クローンについては全長cDNAを得ることが出来た。それらはいずれもウィルスのhypothetical proteinと高い相同性を示し、未知遺伝子と推定されたがその同定にはまだ至っていない。川崎病急性期患児抹消血単核球からは5個のIGHG1クローンを同定した。これらのうち3クローンについての塩基配列をデータベース上に登録されているIGHG1と比較したところ、複数の異なった箇所が見られた。クローン1はC_<H1>領域とC_<H2>領域に1カ所ずつ、クローン2はC_<H1>領域に1カ所、クローン3はC_<H2>領域に3カ所異なった塩基配列を有していた。SEREX解析では二次抗体(患者血清)での反応の前に一次抗体(抗IgG抗体)で処理することでIgGクローンの排除を行っているにもかかわらず、IGHG1が単離されたことはこれらの変異の川崎病患児の免疫系への何らかの関与が推測された。
