Research Abstract

目的:核内へのオリゴヌクレオチド送達を効率化させるナノ粒子を開発し,標的遺伝子の変異修復またはスプライシング反応への介入を目的とする遺伝子治療への基盤技術を確立する.
方法:SV40ラージT抗原由来の核内移行シグナル(NLS)と同一配列のペプチド(-PKKRKV)を付加させたN-イソプロピルアクリルアミド(NIPAA)を化学合成した.NLS-NIPAAとDNA両者を水溶液中で適当な混合比で混和させると径数十nmの複合体(ナノ粒子)を形成する.今回,NLS付加型NIPAAとオリゴヌクレオチド(ONT)を様々な重量比で混合させ,HeLa細胞に導入した.FITCでラベルした25-mer ONTの核内移行効率について蛍光顕微鏡を用いて観察し,2'-O-メチルRNAなどの修飾ONT導入後のアンチセンス(Bcl-xL/SプレmRNAの択一的スプライシング介入)効果についてはRT-PCRを行い,それぞれ市販のリポフェクション試薬との効率の差異を評価した.
結果:窒素/リン酸比0.5-3でFITCラベルONTの細胞内集積が極大を示した.FITCシグナルの核内集積レベル(0.593±0.0706)は,従来のリポフェクション法(0.323±0.0585)を有意に上回っていた(平均値1.83倍;p<0.001).しかしながらBcI-xL/SプレmRNAの択一的スプライシング反応の介入効率については,リポフェクション法が有効であった.
考察:NLS付加型NIPAAはDNAとの親和性が高い一方,細胞内で容易にDNAと解離しないことが今回の結果の原因と考えられた.現在,細胞内環境でDNAと解離する新規の核内送達キャリアーを開発中である.また本研究の過程で得られた,新たな遺伝子導入法に関する知見を国際雑誌に報告した(Sakai et al.,J Biotechnol,2006).
