Research Abstract

ハロタンで麻酔したラットより脳を摘出し,ビブラトームを用いて,酸素93%+炭酸ガス7%で通気し,4℃に冷却したクレブス液内で脳スライス標本(厚さ約150μm)を作成した.ついで,この標本を,酸素93%+炭酸ガス7%(この混合気の通気でクレブス液のpHは7.4となる)で通気し,37℃に加温したクレブス液で満たした観察用チャンバーに入れ,顕微鏡で脳実質内動脈(径約20μm)を観察した.動脈の画像を現有のCCDカメラで撮影し,メディアコンバータを介してコンピュータに取り込んだ動脈径の変化をコンピュータ上で血管径測定用のソフトウエアを用いて解析した.
FHS社製のconcentric bipolar電極(先端径25μm)を用い,0.2Hz,1000pAで脳スライス標本を電気刺激したが,血管は反応しなかった.そこで,同じ電極を用い,刺激強度を400μA,30秒のテタヌス刺激に変更した.この刺激により,脳微小血管は,一過性に収縮した後,拡張反応を示した.この一過性の収縮反応は,ナトリウムチャネル阻害薬であるテトロドトキシン(1μM)の前処置で完全に抑制されたことから,脳内の神経伝達を介する収縮反応であることが明らかとなった.今後,収縮後の拡張反応が,各種K^+チャネル拮抗薬(イベリオトキシン[large-conductanceカルシウム依存性,0.1μM],カリブドトキシン[intermediate-conductanceカルシウム依存性,0.1μM],アパミン[small-conductanceカルシウム依存性,0.1μM],グリベンクラミド[ATP感受性,5μM],4-アミノピリディン[電位依存性,1mM],BaCl_2[内向き整流性,10μM])により修飾されるか否かを検討する予定である.
