Research Abstract

テロメレースはテロメア伸長酵素であり、その活性化は細胞分裂ごとのテロメア短縮を防ぎ、細胞に不死化能を賦与する。テロメレースの活性化はあらゆる癌種に認められる現象であり、癌の分子標的治療のターゲットとなり得る。本研究ではテロメレースを構成する触媒サブユニットであるhTERT (human telomerase reverse transcriptase)をターゲットとして、この遺伝子の発現をRNA interferenceにより抑制し、テロメレース阻害による婦人科癌の遺伝子治療を試みた。
我々はHarvard Medical Schoolとの共同によりhTERT遺伝子発現を効率的に抑制するsiRNA配列を見いだし、これらをretrovirusの系で子宮頚癌細胞(HeLaおよびSiHa細胞)に導入してテロメレース活性の変化および細胞増殖に与える影響を詳細に観察した。siRNA導入により子宮頚癌細胞のテロメレース活性およびhTERTmRNA発現は著明に低下した。これらの細胞はPD40-50で老化を起こし、増殖を停止した。軟寒天培地でのコロニー形成およびマウスでの造腫瘍実験では。siRNA導入で著明な発育抑制が認められ、抗腫瘍効果が明らかとなった。
以上よりhTERTをターゲットとしたsiRNAテクノロジーで癌の増殖をコントロールできることがわかり、今後の臨床応用に向けてさらに研究を継続させる予定である。
