Research Abstract

破骨細胞の分化と同様に,破歯細胞の分化もRANKリガンド(RANKL)とそのおとり受容体であるOPGが調節すると考えられている.OPG遺伝子欠損マウスでは,歯槽骨を含む全身の骨組織において,骨吸収は著明に亢進した状態を呈している.しかし,歯根吸収は全く認められない.そこで,歯根膜組織に存在するOPG以外の歯根吸収抑制因子を明らかにする目的で以下の実験を行った.
1.破歯細胞分化抑制因子の存在様式の解析:OPG遺伝子欠損マウスと野生型マウスの頭蓋骨ならびに切歯歯根より骨芽細胞と歯根膜細胞をそれぞれ単離培養した.これらの細胞と骨髄マクロファージの共存培養系に,RANKLとマクロファージコロニー刺激因子(M-CSF)を添加し,破骨細胞の誘導を試みた.OPG欠損マウス由来の骨芽細胞または歯根膜細胞の共存培養では,野生型マウス由来の細胞を用いた場合に比べて破骨細胞は著明に誘導された.
2.MC-3T3E1細胞が発現する破骨細胞分化阻止因子の解析:マウス骨芽細胞株であるMC-3T3E1細胞は,活性型ビタミンD_3刺激によってRANKLを発現するにもかかわらず,活性型ビタミンD_3による破骨細胞誘導活性を持たないことが報告されている.そこで,破骨細胞前駆細胞とMC-3T3E1細胞を共存培養系にRANKLとM-CSFを添加し,破骨細胞の誘導を試みた.破骨細胞形成はMC-3T3E1細胞数依存的に抑制された.現在,siRNAを使ってOPGをノックダウンしたMC-3T3E1細胞に破骨細胞形成抑制活性があるか検討している.
3.歯根膜周囲組織に発現する遺伝子の解析:マウス歯根膜を含む歯周組織の組織切片を作製し,マイクロダイセクション装置を利用し,歯根周囲歯根膜細胞を回収した.この組織より全RNAを回収した.現在,RT-PCRを用いてRNAの増幅方法を検討中である.
