Research Abstract

1.グリアに発現する神経栄養因子受容体が神経細胞の発生・変性に与える影響の解析
本研究では視覚系の発生に特に重要とされる神経栄養因子BDNFの受容体TrkBに焦点をあて、TrkBのグリア領域特異的コンディショナルノックアウト(CKO)マウスをrdマウス(網膜色素変性症のモデル動物)と交配させ、視細胞変性の進行に与える影響を解析した。昨年度までの研究結果と合わせて、我々のCKOマウスでは視細胞変性の増悪が確認された。また現在は網膜の器官培養系を用いてBDNFによる薬剤誘導性視細胞死の抑制効果を検証中である。
2.培養グリア細胞を用いた神経変性疾患の治療に有用な因子の検討
我々は以前、Muller細胞に神経栄養因子等を投与すると細胞内の栄養因子産生量に変化が起き、周囲の神経細胞の生死や自らの増殖能をコントロールすることを発表した(Neuron,2000)。そこで本研究では神経幹細胞あるいは前駆細胞の増殖や分化に関わる複数の因子を培養Muller細胞に投与・過剰発現、あるいはアンチセンスなどで抑制し、細胞機能の変化を確認している。またこれらのMuller細胞と網膜神経節細胞の共培養で神経保護効果を検討中である。
3.グリアの再生能を利用した神経変性疾患に対する新しい治療法の開発
これまでの研究で我々は、網膜の発生過程においてbasic helix-loop-helix型転写因子の一つであるolig2が分裂能を持つ未分化な網膜前駆細胞に発現することを明らかにした(Neuro Report,2006)。現在はMuller細胞にolig2を過剰発現させ、再生能を腑活化することで神経細胞への分化誘導の可能性を検討中である。これにBDNF・bFGF他の栄養因子等を組み合わせ、特に視細胞と網膜神経節細胞の再生を目指している。
