Research Abstract

前年度までにカチオン性ペプチドとDNAの表面からなるポリプレックスの表面にアニオン性のデンドリマーフタロシアニン(DPc)を配置した光応答型遺伝子キャリアを構築し、動物実験において光照射による遺伝子導入部位の制御に世界で初めて成功した。本年度は、このようなシステムを全身投与による光選択的遺伝子導入に応用するために、ABC型トリブロック共重合体とDNAを混合することにより形成される3層高分子ミセル(JACS 127:2810(2005))のカチオン性のポリマーからなる中間層にDPcを搭載し、表面がポリエチレングリコール(PEG)で覆われた光応答型高分子ミセルを調製した。光応答型高分子ミセルは、90nmの単分散な粒径分布と有しており、DPcの搭載によってアルブミン存在下でのコロイド安定性が高まることが確認された。これは、DPcを搭載することによってアニオン性の中間層が形成され、静電反発によってアルブミンとの相互作用が抑制されることに起因するものと考えられる。DPcは、pH7.4においては中間層に安定に保持されるが、pH5.5においてはDPcが疎水化することによって3層高分子ミセルからリリースされることが示唆された。従って、本システムは、標的細胞に取り込まれた後、エンドソーム内でDPcをリリースし、エンドソーム膜に選択的な光障害を惹起することによって光誘起的な遺伝子発現を示すものと考えられる。実際に、培養がん細胞に対する遺伝子導入効率を評価したところ、光応答型高分子ミセル、光照射によって、細胞毒性を示すことなく、100倍以上のレポーター遺伝子の発現活性を示すことが確認された。本システムは、固形がんへの光選択的遺伝子導入を達成することを目標として、現在、担がんマウスを用いた動物実験を行っているところである。
