Research Abstract

昨年度までの研究に引き続き、初期蛍光が極めて低く、かつ少ないUncaging照射量で大きな蛍光回復の見られる高効率Caged蛍光団の開発を行った。PeT過程の最適化を行い、最終的に2-Me-4-OCH2COOH TokyoGreenをニトロベンジル基でCageした化合物の開発に成功した。このCaged蛍光団は、光照射による蛍光上昇度がほぽ1000倍に達する極めて高効率なものであり、従来のcagedフルオレセインに比べてuncaging光照射量が格段に少なくて済むことが確認された。次に、本化合物のカルボキシ基をAM化した誘導体を合成し、これを生細胞系に適用し、多数ある生細胞のうち1細胞だけを蛍光ラベル化する実験を試みた。その結果、1 microM程度の少量を細胞に負荷し、ここに355nmのuncaging光を10秒程度照射するだけで、照射した細胞だけを十分に明るくラベル化することが可能であることが明らかとなった。従来のcagedフルオレセインでは、同条件下600秒照射してもほとんど蛍光が観測されなかったことから、今回開発に成功したcaged蛍光団の有用性が示された。次に有機合成における効率的光分解性保護基の開発に向け、有機溶媒中で機能するBODIPYべ一スの光分解性保護基の設計・開発を行った。具体的には、BODIPY4位のホウ素上の置換基を種々変換した化合物を合成し、その光分解性を精査した。その結果、4-ベンジル体が500nm程度の強い光照射により開裂することが明らかとなった。本化合物は、分子内にPeT過程を組み込むことで条件判断能を付与することも可能であり、特定条件下でのみ保護基の光脱離が起きうる高機能型光分解性保護基の開発につながる重要な骨格である。そこでまず4位置換基によるBODIPY蛍光の制御が可能かどうかを調べる目的で、酸性条件を認識する蛍光プローブを設計・合成した。その結果、十分な蛍光On/Offが観測され、酸性条件でのみ光脱離する保護基開発の端緒となる重要な知見を得ることに成功した。
