Research Abstract

本研究の目的は精子幹細胞において相同組み替え現象を証明し、その細胞から個体化を行うことである。本年度我々はマウス精子幹細胞(Germline stem, GS細胞)に遺伝子導入を行い、マウスオクルジン遺伝子を破壊するベクターを用いて、オクルジン遺伝子の破壊マウスの作成に成功した。この結果、相同組み替えの頻度は2.3%であり、これは現在幅広く行われているマウスのEmbryonic stem (ES)細胞を用いた場合の相同組み替えの結果(4.5%)に対してやや低いものの十分に精子幹細胞を用いても遺伝子ノックアウトマウスの作成が可能であることを示すはじめての結果である。実際には我々の実験結果は遺伝子導入されたDNAが129マウスの由来で細胞がDBA2マウスだったために、この効率は同じ系統のDNAを使っていれば5-10倍の効率の改善が可能であったことが見込まれており、精子幹細胞はES細胞を利用した場合と少なくとも同等か、それ以上の頻度で相同組み替えを起こすことが可能であることを示すものでもある。
またさらに我々は異種の精巣を使い、ラットの産仔をマウスを仮親にして作成することにも成功した。これによりラットの飼育スペースの作成とその子孫作成にかかる時間を短縮(通常は3ケ月かかるところを6週間に短縮)することに成功した。
この両実験結果はともにProc Natl Acad Sci USA誌のNews articleとして取り上げられ、国内外の報道機関で幅広く報道されるに至った。現在我々のグループはこのマウスでの結果をほかの動物種(特にラット)へと展開し、これまで不可能であったマウス以外の動物種におけるノックアウト動物作成を次の目標として実験を展開している。
